【Pharmacologic effects】

Pharmacodynamics

Ivermectin has good repellent effect on nematodes and surface arthropods. The mechanism of its repellent action is to promote the release of gamma-aminobutyric acid (GABA) from presynaptic neurons, thus opening GABA-mediated chloride ion channels. Ivermectin also has selectivity and high affinity for glutamate-mediated chloride ion channels located near GABA-mediated sites in invertebrate nerve and muscle cells, thus interfering with neuromuscular signal transmission, relaxing and paralysing the worm body, leading to  worm death or expulsion of the body. Inhibitory interneurons and excitatory motor neurons are the sites of action in nematodes, while the sites of action in arthropods are neuromuscular junctions. The elimination rate of Haemonchus, Ostertagia, cooperiasis, trichostrongyles (including trichostrongylus axei), Strongyles, bunostomum, nematodirus spp, ,Trichuris, Dictyocaulus in cattle and sheep， adult Chabertia ovina and fourth stage larvae was 97%-100%. It is also effective against arthropods such as maggots, mites and lice. It is slightly less effective against chewing louse and melophagus ovinus. Ivermectin is also very effective against ticks and flies that breed in their feces. Although the drug does not kill ticks immediately, it can affect feeding, molting and laying eggs, thereby reducing reproductive ability. The effect on blood flies is similar. The elimination rate of Ascaris, Hyostrongylus rubidus, Strongyloides ransomi, Trichomonas elegans, Oesophagostomum, Metastrongylus，Stephanurus dentatus adult and Immature worms in pigs is 94% to 100%, and it is also very effective against trichinella spiralis in the intestine (not effective against trichinella spiralis in the muscle ). It also has good control effect on blood louse and sarcoptes suis. Not effective against flukes and tapeworms.

pharmacokinetics

The pharmacokinetics of ivermectin vary significantly with different animal species, dosage forms and routes of administration. The biological availability of subcutaneous injection is higher than that of internal administration, but the absorption of internal administration is faster than that of subcutaneous injection. After absorption, it can be well distributed to most tissues, but it is not easy to enter cerebrospinal fluid. The apparent volume of distribution of cattle, sheep and pigs is 0.45 ~ 2.4 L/kg, 4.6 L/kg and 4L/kg, respectively. Most animals have long half-lives, 2 to 3 days for cattle, 2 to 7 days for sheep and 0.5 days for pigs. This product is metabolized in the liver, mainly hydroxylated in cattle and sheep, and mainly methylated in pigs. It is mainly excreted in the stool, and less than 5% is excreted in the urine as the original form or metabolite. In lactating females up to 5% of the administered dose is excreted from the milk.

【Drug interaction】  

When used in conjunction with ethylamine, severe or fatal encephalopathy may occur.

【Function and Use】 

Macrolides are antiparasitic drugs. It is used to control nematodes, acariasis and other parasitic insect diseases of domestic animals.

【Usage and Dosage】

Subcutaneous injection: one dose, 0.02 ml per 1kg body weight of cattle and sheep; Pig 0.03ml.

【Adverse reactions】

(1)When used in the treatment of cerercosis, if the larvae killed are in the key parts, it will cause serious adverse reactions.

(2) During the injection, there was discomfort or temporary edema at the injection site.

【Notes】

(1)Cows and sheep that produce milk for human consumption shall not be used during lactation.

(2) Each subcutaneous injection site should not exceed 10ml.

(3)Ivermectin is highly toxic to shrimp, fish and aquatic organisms. Do not contaminate the water source with the remaining medicine and packaging.
